We are currently receiving a large number of applications for data submission, and the review process is taking longer than usual. We sincerely apologize for the delay and kindly ask for your understanding. When submitting an application, we would greatly appreciate it if you could allow sufficient time for the processing.

Following a change to our organizational structure effective April 1, 2026, this division has been renamed from the "Database Center for Life Science, Joint Support-Center for Data Science Research" to the "Database Division for Life Science (DBCLS), BioData Science Initiative (BSI), National Institute of Genetics (NIG)". Where the former name still appears in the guidelines, please read it as the new name.

Research title

Screening for quantitative and qualitative abnormalities of long non-coding RNA associated with gastritis and gastric carcinogenesis

Research overview

Aims
This study aimed to identify dysregulations of long non-coding RNA (lncRNA) in gastric cancer, and to reveal molecular functions of lncRNAs associated with gastritis and gastric carcinogenesis. We also aimed to reveal the correlation between clinicopathological characteristics and lncRNA alterations. To this end, we will try to develop predictive markers of gastric cancer risk, which enables prevention and early detection of gastric cancer. We will also try to develop molecular markers which enable us to select treatment strategies for gastric cancer.
Methods
Genomic DNA was extracted from 17 fresh frozen tissues, containing pairs of non-cancerous gastric mucosa and tumor tissues obtained from 8 patients. We designed a custom AmpliSeq panel that targeted 28 genes. The genes were selected 17 lncRNAs which were aberrantly expressed in gastric cancer and 11 oncogenes. Libraries for amplicon sequencing were prepared using the extracted DNAs according to the manufacturer's instructions. Amplicon sequencing was performed using an Ion Proton sequencer (Thermo Fisher Scientific).
Participants/materials
Non-cancerous gastric mucosa and tumor tissues specimens were obtained from Japanese patients by performing an endoscopic biopsy. Biopsy specimens were nine tumor tissues and eight gastric mucosae obtained from eight patients with gastric cancer.
